"Tools for Exploring Life" PPT

Part One: Situation Import

The rich and colorful biological world contains infinite mysteries, which are waiting for us to explore. Scientific exploration is inseparable from some instruments.

learning target

1. Explain the basic structure and function of a microscope.

2. Identify several commonly used inquiry tools and describe their uses.

3. Use a microscope to observe the prepared temporary slide specimen.

think about it

What instruments can be used to observe fine structures that cannot be seen with the naked eye?

Biology is a category of natural science. It is not only a science with strong basic theory, but also a science with strong experimental nature. It is extremely important to cultivate practical ability.

Exploring the tools of life PPT, part 2: theme exploration

Explore theme one

Get to know the microscope

eyepiece

Optical part of microscope

Function: magnify object image

Magnification is marked on the lens

objective lens

Optical part of microscope

Function: magnify object image

Magnification is marked on the lens

There are low-magnification objective lenses and high-magnification objective lenses

Stage

Mechanical part of the microscope

Function: place slide specimens

reflector

The lighting part of the microscope

Function: Reflect light

There are two types of plane mirrors and concave mirrors

Explore theme two

Use a microscope

1.Preparation

Place the microscope steadily on the experimental table in front of you, slightly to the left, with the arms facing your body and the lens barrel facing forward (as shown in the picture)

2. Aiming at the light ①

Turn the converter to align the low-magnification lens with the light aperture, and turn the coarse focus screw so that the front end of the low-magnification lens is about 1 to 2 cm away from the stage.

To the light ②

Enlarge the aperture;

Keep your left eye looking into the eyepiece while turning the reflector toward the light source with your hand;

Adjust the reflector until you see a bright white circular field of view through the eyepiece. (as the picture shows)

3. Low magnification observation ①

Place the slide specimen on the stage so that the part to be inspected is located in the center of the light hole, and press both ends of the slide with slide clamps. (as the picture shows)

Low magnification observation ②

Watch from the side with both eyes, and stop when the objective lens is lowered to about 0.5 cm away from the specimen. (as the picture shows)

Low magnification observation ③

Use your left eye to look into the eyepiece, turn the coarse focus screw at the same time, slowly raise the lens barrel until the object image appears in the field of view, and then adjust it with the fine focus screw. (as the picture shows)

Precautions

1. When picking up and delivering the microscope, hold the mirror arm with your right hand and the mirror base with your left hand, and handle it gently. Never lift it diagonally or swing it back and forth with one hand to prevent the eyepiece from slipping out and falling.

2. To wipe dust or dirt on the eyepiece, objective lens and reflector, special lens cleaning paper must be used. Never wipe with fingers, handkerchiefs, gauze and plain paper.

3. A converter must be used to switch lenses to avoid bending the optical axis.

4. During microscopic examination, if necessary, tilt the lens tube. Note that the tilt angle cannot exceed 4° to avoid the center of gravity of the entire lens being unstable and causing it to tip over.

5. When turning the coarse and fine focus screws, do not use excessive force to prevent damage to the machine parts and adjustment failure.

Exploring the tools of life PPT, the third part: Class summary

1. Understand the microscope

The product of the magnification of the eyepiece and the objective lens is the magnification of the object image.

2. Use a microscope

Take the lens and place it→Align the light→Place the slide specimen→Observe→Clean and close the lens

3. Other commonly used research tools

4. Preparation of temporary slide specimens

Wipe → drip → collect material → cover → stain → absorb water

Tools for exploring life PPT, part 4: training to meet standards

1. Multiple choice questions

1. Materials to be observed with a microscope must be ( )

A. fresh b. complete

C. Thin and transparent D. dry

2. When observing small objects with a microscope, the eyepiece is marked with "5×". The objective lens is marked "45×". The actual magnification of the observed object is ( )

A. 5 times B. 45 times C. 50 times D. 225 times

3. When you put the slide with "p" engraved on it under a microscope, what you see in your field of view should be ( )

A. q B. p C. d D. b

4. If you write "→" on the slide, it can be seen under the microscope ( )

A. → B. ← C. ↑D. ↓

5. The main steps for making temporary slide specimens can be simplified to ( )

A. Wipe the piece → drip water → collect materials → absorb water → observe

B. Wipe the piece → drip water → take material → absorb water → cover the piece

C. Wipe the slide → drip water → collect material → cover slide → observe

D. Wipe the slide→take the material→absorb water→cover the slide→observe
