Description
"Practice using a microscope" Cells are the basic unit of life activities PPT courseware 4
Self-study guide one
1. Refer to textbook P37 to name and function each part of the microscope.
What is the magnification of a microscope?
The magnification of the microscope = the magnification of the objective lens × the magnification of the eyepiece
Self-study guide 2 (read P37-39 and complete the following steps)
Removing and placing the microscope: Take the microscope out of the mirror box, hold the mirror arm with one hand, support the mirror base with the other hand, and place it about (7) cm away from the edge of the experimental table. Install the eyepiece and objective lens.
Aiming the light: Turn the (converter) so that the (low) magnification objective lens is facing the light aperture, and align a larger aperture with the light aperture. Look at the (eye)piece with one eye and turn the (reflector) until you see a bright, white, circular field of view.
Observation: The specimen is placed on the stage and pressed, facing the center (light hole). Turn the (coarse) focus screw to bring the tube (down) to the objective close to the specimen. At this time, the eyes should be looking at the (objective) mirror. Then, turn the (coarse) focusing screw counterclockwise to slowly (rise) the lens barrel until you can see the object image clearly. Then turn the focus screw slightly (fine) to make the object image you see clearer. Finally, turn the converter to replace the (high) magnification lens, and adjust the (fine) focus screw until you can see the object image clearly.
Cleaning and closing: After the experiment, wipe the surface of the microscope clean. Turn the converter, tilt the two objective lenses to both sides, and slowly lower the lens barrel to the lowest position. Finally, put the microscope into the mirror box.
Learn to use a microscope
Read pages 37 to 39 of the textbook to learn how to use the microscope.
1. Do you have any questions about the operating essentials of a microscope?
2. What are the basic steps for using a microscope?
Take the lens and place it → Aim the light → Observe → Clean and close the lens
Can anyone tell me where this classmate went wrong?
Observe with your left eye.
The microscope should be placed about 7cm from the edge of the experimental table, slightly to the left.
The reflector is not aligned with the light.
After the experiment, organize the experimental equipment
1. Take out the slide.
2. Turn the converter and move the two low-power lenses to both sides.
3. Turn the coarse focus screw to slowly lower the lens barrel and return it to its original position.
4. Place the reflector upright to reduce dust falling on it.
5. Put the microscope into the mirror box and put the mirror box back into place.
6. Place other supplies on the table neatly as they are, and keep the experimental table neat and clean.
Experiment with questions:
1. Can everything be observed with a microscope? Why?
2. How does the word "e" on the slide specimen appear in the microscope field of view? What does this indicate about the characteristics of microscopic imaging?
3. If the specimen to be observed is in the upper left corner of the field of view and you want to move it to the center of the field of view, how should you move the slide specimen?
4. Practice questions 1 to 6 on page P40.
Typical exercises
1. A microscope is a precision magnifying instrument used in junior high school biology learning. When operating the microscope, if the object image is not clear, you should generally adjust (D)
A. Reflector B. Shutter
C. Coarse focus spiral D. Fine focus spiral
2. When observing the temporary mounting slide with a microscope, it was found that there was obvious dirt in the field of view. The dirt did not move when the slide was moved, and the dirt did not move when the eyepiece was rotated. This means that the dirt was on (B)
A. Eyepiece B. Objective lens C. Reflector D. Temporary film mounting
3. Which of the following statements about the use of microscopes is correct (D)
A. When aiming at the light, use a high-power lens to face the light hole
B. To brighten the field of view, replace the low-power lens with a high-power lens
C. Lower the lens barrel and look at the eyepiece when the objective lens is close to the slide.
D. To make the object image you just observed clearer, you can adjust the fine focus screw.
